PSVIII-20 Patatin-Like Phospholipase Domain-Containing Protein 3 Abundance is Affected by Knockdown of Sterol Regulatory Element-Binding Protein 1c but not of Carbohydrate Response Element Binding Protein

Abstract Patatin-like phospholipase domain-containing protein 3 (PNPLA3) is a key lipase involved in fatty liver syndrome onset and recovery humans bovine. Regulation of PNPLA3 mediated by carbohydrate response element binding (ChREBP) rodents sterol regulatory element-binding 1c (SREBP1c) humans. The objectives our study were to optimize an siRNA knockdown ChREBP SREBP1c determine if either transcription factor would affect abundance immortalized bovine hepatocyte cell line, BFH12. Hepatocytes cultured monolayer at least 80% confluent after 24h when cells randomly assigned triplicate treatment with ChREBP-targeted (siC1, siC2, siC3) or SREBP1c-targeted (siS1, siS2, siS3) 20, 60, 80µM, nonspecific-siRNA control (siNON). Media was replenished 4h incubated for 20h before harvest TRIzol subsequent isolation. Protein abundances determined Western blot normalized total lane protein.
